Autoantigen

An autoantigen is a normal protein or protein complex found within the body that, under certain circumstances, becomes the target of an immune response. This aberrant immune reaction can lead to the development of autoimmune diseases. The immune system, typically trained to recognize and tolerate self-antigens, mistakenly identifies autoantigens as foreign invaders, triggering an attack that damages the body's own tissues and organs. The precise mechanisms that lead to autoantigen recognition are complex and not fully understood.

• In Systemic Lupus Erythematosus (SLE), autoantibodies are generated against numerous autoantigens like double-stranded DNA and nuclear proteins. These antibodies form immune complexes that deposit in tissues, causing inflammation and organ damage, resulting in symptoms like fatigue, joint pain, and skin rashes. The variety of autoantigens targeted reflects SLE's diverse clinical presentation.
• Type 1 diabetes is characterized by the autoimmune destruction of pancreatic beta cells, which express autoantigens involved in insulin production. This immune attack, often triggered by environmental factors in genetically susceptible individuals, leads to insulin deficiency and elevated blood glucose levels, necessitating lifelong insulin therapy.
• In Rheumatoid Arthritis (RA), autoantigens present in the synovial fluid are targeted by the immune system, leading to chronic inflammation of the joints. Antibodies against citrullinated proteins and other autoantigens contribute to joint swelling, pain, and eventually, cartilage and bone destruction, causing significant disability.
• Multiple Sclerosis (MS) involves an autoimmune attack on myelin, the protective sheath around nerve fibers.
